The amount or concentration of a drug needed to produce an effect of a given intensity, usually expressed as the EC50 or ED50; the lower the value, the more potent the drug.

Potency is determined by affinity and efficacy together with tissue factors such as receptor density and signal amplification and, in vivo, by pharmacokinetic factors. On a log dose–response curve, a more potent drug lies further to the left; potency is often reported as pEC50, the negative logarithm of the molar EC50.

Potency alone has limited clinical importance: it determines the size of the dose, not the maximal benefit. A more potent drug is not necessarily superior if a less potent one can be given in a convenient, well-tolerated dose. High potency matters mainly for routes with limited capacity (transdermal, inhaled, intrathecal) and in toxicology.

Equi-effective doses differ between drugs of the same class because of potency (e.g. fentanyl vs morphine, bumetanide vs furosemide), so equivalence tables, not milligram-for-milligram substitution, must guide switching.

Daha az potent bir ilaç uygun ve iyi tolere edilen bir dozda verilebiliyorsa, daha potent bir ilaç mutlaka üstün değildir. Yüksek potens esas olarak kapasitesi sınırlı uygulama yollarında (transdermal, inhalasyon, intratekal) ve toksikolojide önem taşır.

Aynı sınıftaki ilaçların eşdeğer etkili dozları potens farkı nedeniyle farklıdır (örneğin fentanil ile morfin, bumetanid ile furosemid); bu nedenle ilaç değişiminde miligram miligram eşleştirme değil, eşdeğerlik tabloları esas alınmalıdır.
